Determination of dissociation constants by affinity electrophoresis: Complexes between human serum proteins and concanavalin A

The general equation for determination of dissociation constants by affinity electrophoresis is given. Dissociation constants of macromolecular complexes may be determined irrespectively of the mobility of the complex. Complexes between two macromolecular components may be studies. Dissociation constants are given for concanavalin A‐complexes of human serum proteins: two components of alpha1‐antitrypsin, two components of orosomucoid (alpha1‐acid glycoprotein), high density lipoprotein‐associated arylesterase, haptoglobin, alpha2‐HS‐glycoprotein, and transferrin. When the affinity electrophoresis is performed as lectin interaction electroimmuno precipitation (crossed immunoelectrophoresis with lectin in the first dimension gel) prior purification is unnecessary.
