Pharmacokinetics ofAzlocillin inSubjects withNormaland Impaired RenalFunction

half-life (T1/2a) was0.11h,andthemeanelimination serumhalf-life (T12p) was 0.89 h.Thevolume ofthecentral compartment (Vc)was7.36 liters/1.73 m2,and theapparent volume ofdistribution (Vd.) was14.15 liters/1.73 m2,i.e., 21.9% of bodyweight. TheTI/,B after a30-niin intravenous infusion of80mg/kgtothe samehealthy subjects was1.11h.Serumclearances (Cs) forthe30-and80-mg/ kgdoses were215.0 and152.9 ml/min per1.73M2.Themeanrenal clearances (CR)were145.2 and94.1ml/min per1.73m2fortherespective doses. Between 61.8and69.6% oftheinjected dosewasrecovered inurine during thefirst 24h. Theelimination half-life insubjects withchronic renal impairment increased with thedegree ofrenal insufficiency. After a30-min intravenous infusion of80mg/kg theTI/2, values were2.03, 4.01, and5.66 hwithcreatinine clearances (C(r) within 30to50,10to30,and<10ml/min per1.73 m2,respectively. Urinary elimination wasinversely related tothedegree ofrenal impairment. Infourpatients outof andona6-hhemodialysis session meanelimination half-life values were6.53 and 2.81h,respectively. Thefraction ofdrugremoved bydialysis was45.8%. The linear relationships between theelimination ofhalf-life (T1/2p) andserumcreatinineandtheelimination rateconstant (,B) andcreatinine clearance (Ccr) provided abasis foradjustment ofdosage inrenal failure. Azlocillin, aparenteral semisynthetic penicillin, oneofagroupofacylureidopenicillins, has a broadspectrum ofactivity covering non-f,lactamase-producing
