pML31, a plasmid chimera constructed tocontain thereplication genesofan Flacplasmid, hasbeenstudied byelectron microscope methods. Heteroduplex analysis showsthattheonlyF sequencepresent inpML31isthatwithcoordinates40.3-49.3F. Thisregion haspreviously beenidentified as essential for plasmid maintenance. ThesequenceofpML31,which was derived originally fromR6-5, carries thekm gene(s) andan inverted duplication ofa 1.0-kilobase sequence.On thebasis oflength measurements, therepeated sequenceis different fromIS1,IS2,IS3,andan inverted repeatassociated withthekm gene(s) ofplasmid JR67.
