The invention discloses a route type upward horizontal layered filling mining method suitable for extremely complex ore body mining. The method comprises the following steps of: performing ore block division on an ore body, reserving a bottom pillar with height of 8 meters at the bottom of the ore body, forming an air return tunnel at the top of the ore body, and performing development construction, namely forming a transport tunnel in the bottom pillar and upwards forming an equipment well, a man-way shaft and an ore winze communicated to a stope along the transport tunnel respectively. The method is characterized in that: the lower plate of the ore body of a mining area is flush with the ore body and advanced with an auxiliary operation tunnel with width of 4 meters and height of 2.8 meters together with the transport tunnel; after the construction is finished, a ventilating patio is upwards filled and communicated with the air return tunnel; and then mining construction is performed, wherein the mining construction comprises equal distance cross tunneling of a primary route and a secondary route, upward recovery and cross filling till the top of the ore body is mined. According to the method, the advantages of a traditional upward horizontal filling mining method and a traditional upward route filling mining method are integrated, and ore resources can be reclaimed with low cost under extremely complex geological conditions; and the method is particularly suitable for mining the mines of copper or iron and noble metals.
