Metabolizing Systems Used for in Vitro Mutagenicity Testing

In early 1977 a workshop was held to discuss the plate incorporation assay (Ames test) to detect mutagenic effects of chemicals (1–3). The aim of the workshop was to discuss a protocol and to identify those parts of the procedure which should be defined better. It turned out that the system for metabolic activation of the test compounds especially required better definition and standardization. All details were discussed and a revised test protocol has been elaborated (4). A consensus was reached that no rigid protocol should be prepared because the users of the Ames test are rather to be encouraged to vary the test procedure to adapt it to the special requirements of the different test compounds.
