Supramolecular Approaches to Medicinal Chemistry

This chapter describes how the central concept of molecular recognition links the distinct pursuits of supramolecular chemistry and drug development. Topics covered include the use of enzyme inhibitors as tools for measuring the strengths of weak interactions, the use of drug targets as templates for the formation of their own inhibitors (both in situ templated synthesis and dynamic combinatorial chemistry (DCC)), and the use of traditionally supramolecular scaffolds for the recognition of large protein surfaces. Keywords: drug–target interactions; dynamic combinatorial chemistry; multivalency; protein interactions; templated synthesis; weak interactions
