The Varifocal Scanning Micro-Zooming Technique: A New Method for Measuring the Neurite Length in Three Dimensional Spaces

Neurite length measurement is very important in describing neuron's physiological states and of morphological parameters. Combining computer technology and modern microscopic optical technology can develop a high precision measurement for measuring the neurite length in three-dimensional space. Through record Zeiss microscope spiral rotation angle in two different planes and Matlabs excellent image process compatibility, the neurite total length and the distance between two adjacent plane can be stored and analysised. Single and connect centerlines alone neurite are extracted. Based on the fast neurite centerlines extracting systems and automatic data processing, we have produced an automated neurite labeling and measuring system that could be used for quantitatively studying neuronal structure in 3D cultures.
