Spectrophotometric Determination of Polysaccharide in Pinus Seed Shell with Ultrasonic Extraction

Objective This paper studied the ultrasonic hot water extraction of the polysaccharide from Pinus seed shell. Methods The effects of four factors such as the temperature of extraction,the time of extraction,the proportion of water volume,the number of extraction on the extraction were investigated. Results and Conclusion With the orthogonal design of L9( 34),the optimum conditions of extraction were obtained as follows: temperature 60℃,Time 25 min,ratio of solid- liquid 1: 3. 5,extraction twice. Tests for recovery were made by standard addition method,values of recovery found( n = 5) were in the range of 97. 71% ~ 100. 67% and value of RSD was 1. 56%. Based on these factors,a easy and efficient method of sepectrophotometric determination of polysaccharide in Pinus seed shell with ultrasonic extraction.
