Validation of Transgenerational Otolith Marking Using Alizarin Complexone for Embryos in Black Rockfish Sebastes schlegelii Ovaries

Transgenerational marking method for otoliths of embryos in pregnant viviparous fish is described. Pregnant female parents of the black rockfish, Sebastes schlegelii, were immersed in alizarin complexone (ALC) solution for 30 hr. The delivered larvae were cultured. Then the otoliths (sagittae and lapilli) of the larvae and juveniles were observed at 0, 5, 35, 55, and 110 days after birth (DAB) using UV light microscopy. Markings on lapilli were detected until 110 DAB, about TL 90 mm. Results from this study indicate the effectiveness of ALC marking for embryos in ovaries of the black rockfish.
