genetic analysis: Selection and amplification of rare functional nucleic acids

This article describes methods for the in vitro selection and amplification of rare functional nucleic acid molecules. An overview of strategies and applications for pool construction, selection, and amplification is presented. Techniques and precautions are discussed in the context of two specific selection systems: In the first system, functional ribozymes that can catalyze a ligation reaction are selected from pools of largely nonfunctional molecules. In the second system, RNA molecules that can bind to affinity columns are selected from random sequence pools.
