BsmI polymorphism in N-ras intron I (RASN).

Source/Description: Oligonucleotide primers were used to amplify a region of N-ras, including sequence from intron I and exon II. Polymorphism: BsmI identifies a simple two allele polymorphism with bands at 370 bp or 110 and 260 bp. Frequency: Studied in 68 unrelated individuals allele Fl (360 bp) 0.61 allele F2 (110 and 260 bp) 0.39 Chromosomal Location: N-ras has been localized to Ipl3 (1). Mendelian Inheritance: Co-dominant segregation demonstrated in two families (11 individuals). PCR Conditions: 1 /tg of genomic DNA was amplified using 1 /tg of each primer, 2 units of Taq polymerase and the buffer provided (BCL). After heating at 97 °C for 5 min enzyme was added and samples were cycled thirty times at 94 °C, 61 °C and 74°C, allowing 1 min at each temperature.
