The macromolecular organization of the elastin fibril.

Insoluble elastin was isolated from bovine ligamentum nuchae by treatment of the tissue with guanidine hydrochloride and dithiothreitol, followed by digestion with collagenase purified by affinity chromatography. Under the electron microscope negatively stained elastin fibrils were resolved into primary filaments arranged in a lateral array of about 50 A periodicity. The anisotropy of the preparation was also indicated by optical polarization analyses and by the presence of equatorial reflections in the low-angle X-ray diffraction patterns of stretched specimens. The wide-angle X-ray diffraction patterns of stretched and relaxed specimens showed only two broad diffraction rings.
