Information Processing in Digital Holographic Microscopy

Digital holography is a new three-dimensional imaging technology. The recording conditions of hologram in off-axis Fresnel transform digital holography with pre-magnification are analyzed. The experimental setup used for recording the digital hologram of microscopic object is built. The numerical reconstruction methods of digital hologram disturbed by strong noise are studied experimentally. The intensity and phase image of the USAF test target is obtained by Fresnel transform algorithm. The lateral resolution of and axial resolution of no larger than 10 nm are achieved. The results show that for strong noisy hologram the automatic phase compensation method must be combined with manual adjusting parameter in order to obtain accurate phase image.
