Cloning and Analyzing the Gene cDNA Sequences of Resistance to Uncinula necator from Vitis pseudoreticulata

To identify and clone powdery mildew resistance related genes,mRNA differential display polymerase chain reaction(DDRT-PCR)was used to analyze the differential expression genes from disease resistance of V.pseudoreticulata clone Baihe-35-1 inoculated with Uncinula necator.Through the mRNA differential display,184 DDRT-PCR primer combinations suitable for grape were screened out and 13 differential cDNA fragments were obtained,which related to transcriptional control,energy metabolism,protein synthesis,defense reaction and the other physiochemical process.The results suggested that V.pseudoreticulata Baihe-35-1 resistance to powdery mildew is the co-effect result of many metabolic pathways.These differential cDNA fragments will provide a theoretical foundation to elucidate the molecular mechanisms of Vitis resistance to powdery mildew.
