İneklerde Seksüel Siklusun Farklı Dönemlerinde Uterus Arginaz Seviyelerinin Araştırılması

Bu calisma ineklerde seksuel siklusun farkli donemlerinde uterus arginaz seviyelerinin belirlenmesi amaciyla yapildi. Calismanin materyalini yaslari 3-7 arasinda degisen, farkli irktan toplam 40 inek uterusu olusturdu. Uterus ornekleri duzenli olarak kesim yapilan yerel bir mezbahadan temin edildi. Kesimden hemen sonra genital organlar inspeksiyonla muayene edilerek ic genital organlarinda herhangi bir patolojik lezyon gozlenmeyen hayvanlara ait uterus ornekleri calismaya dahil edildi. Hayvanlarin seksuel siklus donemi, ovaryumlar ve uterus muayene edilerek belirlendi. Uterus doku arginaz aktivitesi Tiyosemikarbazid-Diasetil Monoksim Ure (TDMU) metodu ile spektrofotometrik olarak olculdu. Uterus arginaz duzeyleri proostrus doneminde 0.56±0.19 U/mg protein, ostrus doneminde 0.68±0.23 U/mg protein, metostrus doneminde 0.59±0.21 U/mg protein, diostrus doneminde ise 0.50±0.12 U/mg protein olarak belirlendi. Elde edilen degerler arasindaki farkin istatistiksel olarak onemli olmadigi (P>0.05), ancak degerlerin seksuel siklus donemlerine gore sayisal farkliliklar gosterdigi tespit edildi. Ozellikle ostrus doneminde doku arginaz duzeyinin yukseldigi gozlendi. Sonuc olarak, ineklerde ostrus siklusunun farkli donemlerinde uterus dokusu arginaz seviyeleri arasinda bir farkliligin olmadigi goruldu. SUMMARY The aim of this study was to investigate arginas status in different sexual stages in cattle uterus. Forty, 3-7 years old, different breed cow's uterus were used in the study. Uterine samples were obtained from cows slaughtered routinely at the local abattoir. Immediately after slaughter, uterus and ovaries were visually examined to determine any pathologic lesions of the genital tract and uterine samples were dissected only from cows that were diagnosed to have a healthy genital tract. The sexual cycle of cows was diagnosed via examination of ovaria and uterus and divided four equal groups. The thiosemicarbazide- diacetylmonoxime urea (TDMU) method was used to measure uterus arginase activity. Uterus arginase levels (U/mg protein) in proestrous, estrous, metestrous and diestrous was 0.56±0.19, 0.68±0.23, 0.59±0.21, 0.50±0.12 U/mg protein respectively. No statistically significant difference was observed uterus arginase levels in different sexual stages of cow's estrous cycle. As a result, cow uterine arginase levels were not changed different sexual stages in cows.
