Characterization of an Isolate of Cucumber mosaic virus from Raphanus sativus L.

Choon-hyang), whichshowed mosaic symptoms. The isolate was confirmed to a strain of CMV by host responses in Vignaunguiculata, Chenopodium amaranticolor and Gomphrena globosa, by viral genome composition with RT-PCR and PCR-RFLP, and by serological analysis. Symptom developed by the strain of CMV was severe inNicotiana benthamiana, N. glutinosa, N. tabacum (cv. Samsun, cv. Xanthi), Cucumis melo (cv. Early hanover),Cucumis sativus (cv. White wonder), Capsicum annuum (cv. Chung-yang and cv. Geum-top), but mildsymptom was developed in Raphanus sativus (cv. Choon-hyang), Brassica rapa ssp. pekinensis (cv. Bul-AmNo. 3), and B. juncea (cv. Daenong Jukgot). Newly isolated strain of CMV could infect diverse crops includingSolanaceae, Cucurbitaceae and Brassicaceae. We designated the new strain of CMV as Gn-CMV based on thenovel infectivity of Brassicaceae. In double-stranded (ds) RNA analysis, Gn-CMV consisted of 3.3, 3.0, and2.2 kb genomes likewise other strains of CMV. SDS-polyacrylamide gel electrophoresis (SDS-PAGE) showed28 kDa of the CMV coat protein. By restriction enzyme mapping using Cac8I, ClaI and MspI of RT-PCRproducts indicated that Gn-CMV belongs to CMV subgroup I. Keywords : dsRNA, Host range analysis, PCR-RFLP, RT-PCR, SDS-PAGECucumber mosaic virus(CMV)는 Bromoviridae Cucumovirus에 속하며, 직경 29 nm의 구형으로 되어 있고, 박과, 가지과 등 85개과의 1000종 이상의 넓은 기주범위를 갖는다(Palukaitis 등, 1992). 전 세계적으로 널리 분포하고 많은 종류의 작물에 피해를 주고 있는 CMV는 채소나 화훼에서는 가장 중요한 바이러스병의 병원이다(Tomlinson,1987). CMV는 게놈 분자의 크기에 따라 RNA1, 2, 3의분절된 genome을 가지며(Palukaitis 등, 1992), 서브게놈RNA(RNA 4)를 포함하고 있다. CMV는 계통에 따라서는 satellite RNA를 함께 갖고 있는 CMV도 있다(Gould등, 1978). 세계적으로 많은 계통과 분리주가 보고되어 있는 CMV는 기주 반응의 특성과 혈청학적 성질, 외피단백질의 peptide 서열과 ELISA 분석, RT-PCR을 이용한 외피단백질 유전자의 제한효소 절단패턴 분석 등에 따라subgroup I과 II로 구분된다(Choi 등, 1998; Owen과Palukaitis, 1988). 또한 RNA3의 외피단백질 유전자와 5’비번역 부위를 통하여 subgroup IA와 IB로 구분하고 있다(Roossinck 등, 1999).무, 배추 등 십자화과 식물에 감염하고 있는 바이러스로는 대표적으로 Potyvirus 속의 Turnip mosaic virus(TuMV)가 보고되어 있으며(Tomlinson 등, 1970), Tobamovirus속의 Ribgrass mosaic virus(RMV; Wang 등, 1997), Turnipvein clearing virus(TVCV; Lartey 등, 1993), Cr-TMV(Dorokhov 등, 1994), Chinese rape mosaic virus(CRMV;Aguilar 등, 1996), TMV-Cg(Yamanaka 등, 1998) 등이 보
