Constitutive andInducible Secretion ofPlatelet-derived GrowthFactor AnalogsbyHuman LeukemicCellLinesCoexpressingErythroid andMegakaryocytic Markers

We haveexamined theconstitutive andinducible secretion of platelet-derived growth factor (PDGF)-like proteins ina variety ofhumanhemopoietic cell lines. Thehighest levels ofsecreted protein were noted infourhumanerythroleukemia lines which, inaddition toerythroid lineage markers, express one or more megakaryocytic lineage markers. Induction ofthese lines by12O-tetradecanoylphorbol-13-acetate enhanced theexpression of megakaryocytic markers andincreased secretion ofPDGF-like proteins several fold. Inconcert withthese changes, there was significant induction ofc-sis/PDGF-B messengerRNA (mRNA) expression inalllines, whereas oneline showedsignificant concurrent induction ofPDGF-A mRNA expression. Whether PDGF-like secretion ispartofthestemcell-like phenotype displayed bythese lines orissecondary totheir leukemic transformation remains tobedetermined. Nevertheless, these lines provide newcellular models forstudying theexpression andfunction ofPDGFanalogs inhemopoietic cells.
