A Non-Linear Model of Insulin Kinetics

A five compartment model including receptor-mediated insulin degradation is proposed to simulate insulin processing during hyperinsulinaemic euglycaemic clamps. Several physiological constraints were employed to limit the number of parameters which remained to be resolved from the experimental data. The model gave estimates of otherwise inaccessible parameters and also predicted the time course of insulin concentration in various physiological compartments. The model was validated by comparing the summarised variables, i.e. receptor association, receptor affinity, basal degradation in kidney, liver and periphery, to corresponding values from the medical literature.
