Synthesis in vitro of high-molecular-weight RNA by isolated Euglena chloroplasts.

Chloroplasts isolated from photoheterotrophically grown Euglena gracilis incorporate [3H]UTP into RNA. This incorporation is sensitive to DNAase, RNAase and actinomycin D and requires the presence of magnesium and the other three nucleoside triphosphates. Analysis of the products of the reaction revealed that this incorporation is due to elongation of RNA molecules, resulting in peaks of newly synthesized RNA coincident with the 23 S and 16 S chloroplast ribosomal RNAs. These findings are consistent with chloroplast ribosomal RNA being synthesized in vitro using chloroplast DNA as a template.
