Progenitor cell kinetics in periodontal tissues in vitro.

歯肉剥離掻爬手術後の新付着成就に, 従来から歯根膜由来細胞の関与が示唆されているが, 新付着形成細胞の起源については, 未だ明確にされていない。そこで本研究では, 新付着形成細胞の起源解明の一助として, 移植片培養したサル歯周組織における細胞動態を検索した。実験動物には, 健康歯肉を有するニホンザルを用い, 臼歯部頬側歯肉から上皮除去後, 歯肉弁を剥離, 歯槽骨を骨頂から根尖方向へ約3mm削除した。次いで, 歯一歯根膜一歯槽骨一歯肉結合組織を一一塊として取り出し, 移植片培養を行った。培養開始後1, 3, 7, 10, 14日目の各移植片を位相差顕微鏡的, 光顕的, 電顕的に観察した。その結果, 移植片内での細胞増殖は骨髄腔で最初に生じることが示された。骨髄腔に続いて, 歯根膜血管周囲組織および歯根膜骨側に細胞増殖が観察された。骨髄腔や歯根膜血管周囲, 歯根膜骨側での活発な細胞増殖に対して, 歯根膜セメント質側では細胞の増殖能は低かった。また, 歯槽骨頂から歯冠側へ派生した細胞が, 歯根膜上を分化しながら根面へ移動したが, 歯根膜から歯冠方向への細胞派生はみられなかった。以上の所見から, 術後, 根面へは, 歯槽骨, 歯根膜血管周囲組織, 歯根膜骨側由来の各細胞が集積するものと考えられ, これらの細胞群が新付着形成に関与するものと思われた。
