Optimization of Ultrasonic Extraction of Phellinus igniarius Polysaccharides Using Response Surface Methodology

Response surface methodology (RSM) was used for optimizing ultrasonic extraction conditions in the extraction of polysaccharides from Phellinus igniarius. Based on single-factor experiments, three independent variables, namely extraction time, solvent-solid ratio and ultrasonic power, were selected as casual factors during extraction. The experiments were arranged according to Box-Behnken central composite experiment design. Response surface analysis was applied to determine the effect of central composite experiment design and the effect of casual factors on the yield of polysaccharides from P. igniarius. The results showed that the optimum ultrasonic extraction conditions for polysaccharides were as follows: material 0.1g, extraction time 260s, ultrasonic power 464W and ratio of liquid to solid 49∶1 (mL∶g). The yield of polysaccharides for twice was 13.19%,which was higher than the traditional extraction with hot water, and extracted time was significantly reduced.
