Abstract The separation of 2′-5′ dinucleoside monophosphates from their corresponding 3′-5′ isomers on QAE-Sephadex A-25 (bicarbonate form) is described. The column is loaded with a mixture of the two isomers and eluted with a linear gradient of ammonium bicarbonate (from 0.02 to 0.2 M ) The two isomers appear in two different peaks, and the nucleotidic material is isolated by lyophilization. In addition, the separation of 2′-5′ dinucleoside monophosphates from the corresponding 3′-5′ isomers by paper chromatography and by thin-layer chromatography on aluminium cards coated with cellulose powder is described.
