DETECTION OF AMPHOLINE PATTERNS *

The composition of the widely used camer ampholytesla originating from a single commercial source (Ampholirie) is thought to be complex. Estimations for the number of individual ampholytes run up to several hundreds,'* but these estimations are difficult to substantiate. It is also unknown, whether the single ampholytes arc present in comparable concentrations and whether they are distributed evenly over the entire pH range or not. Absorbancy of column eluates at 260 nm may indicate the distribution patterns of ampholytes,22 and Fratero has desaibed a Coomassie violet staining procedure for Ampholines after gel focusing. Accidentally, we discovered that ampholines produce intense brown colors if heated to dryneas with glucose, probably by a caramelization process. After Sephadex thin layer focusing, a reproducible Ampholine pattern can be visualized within minutes.
