Factors that affect the extraction of xyloglucan from the primary cell walls of suspension-cultured rose cells

Abstract The extraction of xyloglucan from Rosa suspension-culture cell walls (not depectinated in hot aqueous solutions) with alkali was dependent on the temperature, with 2–3 times more being extracted at 37° than at 25°. At 25°, 6 m NaOH was the optimal concentration, but it extracted only 25–30% of the xyloglucan within 24 h; efficient extraction took 2–3 weeks. After pre-treatment of cell walls with 6 m NaOH at 25°, water extracted more xyloglucan in 24 h than did additional 6 m NaOH.
