Practical aspects of the calibration and use of the Aminco-Bowman spectrophotofluorometer.

Abstract Although the Aminco-Bowman spectrophotofluorometer is widely employed as an analytical tool, relatively few users of the instrument seem to be familiar with its basic optical features. As a result, optimal performance of the apparatus is frequently not obtained, although it can be adapted to a wide range of fluorescence measurements including corrected excitation and emission spectra, quantum yields, polarization of fluorescence, as well as simple measurements of fluorescence intensity. Increasing interest in emission spectroscopy in recent years has generated a demand for more sophistication in the use of instruments for measuring fluorescence. Fortunately, coincident with these requirements, there have been marked improvements in the stability of lamp sources and in the quality of optical components. It does not seem unreasonable, therefore, that commercially available fluorescence spectrometers may be used for purposes other than the simple measurements for which they were originally intended. The present paper discusses some practical aspects of the use of the Aminco-Bowman spectrophotofluorometer for accurate measurement of spectra in the hope that such a communication will be helpful to those who wish to extend the usefulness of the instrument.
