The invention belongs to the field of molecular genomics and especially relates to the field of a simplified genome sequencing technology. More specifically, the invention relates to a 2b-RAD pooling technology. According to the invention, twenty 2b-RAD pooling barcode connectors and one general connector are designed, wherein the general connector is a SEQ ID No.31 and 32 paring, and SEQ ID No.1-10 are respectively paired to SEQ ID No.21-30 to form ten barcode connectors and the general connector which are used for pooling after BsaXI digestion; and SEQ ID No.11-20 are respectively paired to SEQ ID No.21-30 to form ten barcode connectors and the general connector which are used for pooling after AlfI digestion.
