Conversion of methionine to Xanthomonas campestris pv. phytotoxic 3-methylthiopropionic acid by

Incubation of the plant pathogenic bacterium Xanthomonas campestris pv. mmihotis with either ~-[3,414C]methionine or [3,4- 4C]KMBA (2-keto-4-methylthiobutyric acid) led to the production of [l 4C]MTPA (3methylthiopropionic acid) and [ 4C]MTAA (3methylthioacrylic acid). When an excess of non-radioactive KMBA was present in the medium, formation of [ 14C]KMBA from ~-[3,4-~~C]methionhe was observed. Conversely, [ 14C]methionhe accumulated in a trapping pool of non-radioactive methionhe as a result of biotransformation of [3,4-14C]KMBA. Aminooxyacetic acid, a transaminase inhibitor, suppressed totally the formation of [ 4C]MTPA from ~-[3,4-l 4C]methionine but not from [3,4-' 4C]KMBA. Metabolism of L-[ l-14C]methionine liberated 4C02 but did not produce [ l4C]MTPA. These results demonstrate that methionhe is catabolized by X. campestris pv. mmihotis into MTPA via transamination and subsequent decarboxylation of the intermediate a-keto acid KMBA.
