Histological and Morphometrical Properties of Cattle Epidermal Langerhans Cells

13頭のウシを用い,抗原提示細胞である皮膚表皮ランゲルハンス細胞(L細胞)の組織学および計量形態学的特性を調べた.50ヵ所の皮膚で,L細胞を亜鉛沃素オスミウム(ZIO)法で,組織化学的に特異的に検出した、細胞分布密度(VD)および細胞周囲長/細胞面積比(GAR)を,自動画像解析装置を用いて計測した.その結果,(1) われわれは,0.05Mのベロナール塩酸緩衝液(pH 7,6)で緩衝したZIO溶液で,4°C 18時間染色することによって,ウシL細胞を選択的に染色できることを明らかにした.(2) L細胞を,細胞突起の数(1-3,4-6,7-9および10以上)に対応して,各々をI,II,IIIおよびIV型に分類した.手掌,足底部等の表皮肥厚部においては,大部分の細胞体は,有棘層中央部に局在し,長くて高度に細分岐してしばしば淡明層に達する細胞突起を持っていた(II,IIIおよびIV型).逆に,腋窩,そ径部のような薄い部位では,細胞体は,有棘層底部に局在し,短くて顆粒層細胞底部で終了する細胞突起を持っていた(IおよびII型),一般に,高度にケラチン化した部位のL細胞は,低ケラチン化部位の細胞と比較して,より多数の細胞突起を持つことが判明した.(3) 表皮肥厚部および粘膜の近傍部位においては,VD(1,100細胞/mm3以上)およびGAR(6.00以上)ともに高かった.逆に,耳介,外耳道部等のわずかの例外はあるが,表皮の薄い部位では,VD(500細胞/mm3以下)およびGAR(4.00以上)ともに低かった.加齢に伴ってVDは増加する傾向があったが,GARには変動が認められなかった.細胞の組織学および計量形態学的特性は,皮膚における抗原提示機能に適応していると考えられた.
