A procedure for evaluation of significant structural differences between related arrays of protein molecules

Abstract A computational procedure is presented for the quantitative evaluation of structural differences between related assemblies of protein molecules as represented in filtered electron micrograph images. The procedure, whose criterion for significant differences is based on the reproducibility of detail between different images of identical specimens, is demonstrated in a model experiment and applied to micrographs representing the cleaved and uncleaved states of the bacteriophage T4 prohead capsid.
