RNA m6A demethylase ALKBH5 regulates the development of γδ T cells

Significance γδ T cells are an abundant T cell population that is critical in mucosal tissue homeostasis and immunoregulation, while its detailed developmental mechanisms remain poorly understood. Previous studies have demonstrated N6-methyladenosine (m6A) plays important roles in controlling the homeostasis and differentiation of CD4+ αβ T cells. However, the roles of m6A RNA modification in the development and function of γδ T cells remain unclear. Here, we report that ALKBH5-mediated m6A RNA demethylation regulates the early development of γδ T cell progenitors by specifically fine-tuning the messenger RNA (mRNA) expression of NOTCH signaling genes.
