Induction of IL-2 activity production in human T cell line, KOPT-5

T細胞型急性リンパ性白血病患者末梢血からT細胞株KOPT-5を樹立し,そのIL-2活性産生能を検討した. KOPT-5は, CD 1 (-), CD 2 (+), CD 3 (+), CD 4 (-), CD 5 (+), CD 7 (+), CD 8 (-)で, PMAによりマクロファージ様に分化させたHL-60とPHA存在下に混合培養するとIL-2活性を産生した. KOPT-5によるIL-2活性の産生は,マクロファージ様に分化させたHL-60との混合培養のみでは得られず,また,分化させないHL-60とPHA存在下に混合培養しても得られなかった. KOPT-5をPMAとPHAとで同時に刺激すると,少量のIL-2活性の産生が認められたが, PHAとIL-1による同時刺激ではIL-2活性の産生は全く認められなかった. T細胞の活性化にかかわる種々の因子,特にマクロファージの影響を解析するうえで, KOPT-5は有用なモデルになると考えられる.
