Sectioning Pupae with Sclerotized Cuticle

AbstractThe technique described permits serial sections to be cut from the pupae of Phaormia terranova without having to remove the epidermis before fixation. The pupae were prefixed for 1 hr at high temperature (85°C) in Bouin fluid. Their cuticulae were then removed before the pupae were postfixed at room temperature in Bouin fluid. The correct topography of the tissues was retained. (The J Histotechnol 14:49, 1991)
