Cloning of a coding sequence of "Bordetella pertussis" filamentous hemagglutinin gene.

An expression library of Bordetella pertussis DNA sequences obtained by DNAase I digestion was constructed in bacteriophage λgt11. A clone with an insert of 2.9 kb which expresses epitopes recognized by B. pertussis filamentous hemagglutinin (FHA) antibodies was isolated. FHA antibodies immunopurified using immobilized β-galactosidase-FHA fusion protein detect specifically FHA in B. pertussis culture supernatants on Western blots. A partial sequence of the λgt11 DNA insert reveals a 65% GC content of the gene.
