Chromosomal assignment of genes in barley using telosomic wheat–barley addition lines

Characterized DNA clones have been used as probes in Southern blotting and hybridization analysis of genomic DNA from a set of wheat–barley telosomic addition lines to assign genes to chromosome arms in barley (Hordeum vulgare). The lines used included a newly available addition line with the short arm of chromosome 5. Addition lines with the long arm or the whole of chromosome 5 of H. vulgare are not available, so addition lines with chromosomes 5 and 5S of a related wild species, Hordeum chilense, were also used. A total of 23 characterized cDNAs and two characterized genomic clones were mapped to chromosome arms in this way.Key words: gene mapping, telosomic additions, cDNA clones, wheat-barley, restriction fragment length polymorphisms.
