EFFECT OF PINEAPPLE AND POMEGRANATE JUICES ON CYP2C9 MEDIATED GLIMEPIRIDE METABOLISM INVITRO

The objective of this study was to determine the effect of pineapple and pomegranate juices on CYP2C9 mediated metabolism of glimepiride invitro using human liver microsomes. The addition of 10 µl (5% v/v) of pineapple juice resulted in almost complete inhibition. On the other hand pomegranate juice had less CYP2C9 inhibitory capacity. At concentrations 0.5% v/v the percentage inhibition was 60.34% and at 1.5% v/v it was 29.42% for pineapple juice. Similarly for pomegranate juice, at concentrations 0.5% v/v the percentage pineapple juice (PIJ) pomegranate juice(POJ) the CYP2C9-mediated drug fruit interaction with glimepiride. glimepiride as a model substrate with pineapple and pomegranate juices. of microsomal protein. The concentration of glimepiride was 10µM. Tubes (duplicate) containing the reaction mixture in phosphate buffer and NADPH solution (10mM) were allowed to equilibrate in a shaker incubator at 150rpm for 5 minutes at 37°C. Preliminary experiments showed that the substrate depletion was linear with respect to time over 30 min and liver microsomal protein concentration of 0.5mg/ml at 37°C. The reaction was initiated by addition of NADPH (preincubated for 5 min) and terminated with 100  l ice cold acetonitrile. The tubes were centrifuged at 10,000 rpm (4°C;10min) and aliquots of the supernatant were directly injected into an HPLC system. Wherever necessary volume was made upto 200  l with buffer.
