Background: Strain and estrogens down-regulate Sost /sclerostin and stimulate osteoblastic proliferation. Results: ER (cid:1) inhibition prevents proliferation. ER (cid:2) inhibition prevents Sost down-regulation by strain or estradiol. Sclerostin prevents proliferation following strain and not estradiol. Conclusion: ER (cid:1) promotes proliferation, and ER (cid:2) mediates Sost down-regulation following estradiol ligand stimulation and ligand independently following strain. Significance: Selective ER modulators could promote osteogenesis through differential regulation of Sost and proliferation. Mechanical strain and estrogens both stimulate osteoblast proliferation through estrogen receptor (ER)-mediated effects, and both down-regulate the Wnt antagonist Sost/ sclerostin.
