In vitro regeneration study in lablab bean and dolichos bean (Lablab purpureus (L). Sweet) Genotypes

The current investigation was carried out to study in vitro regeneration potential in Lablab bean genotypes and Dolichos bean Cv. (Konkan Bhushan). In surface sterilization, treatment with 70% alcohol for 5 minute followed by HgCl2 (0.1%) for 10 minutes was found the most effective in establishing aseptic cultures (97.22%). Mature embryo axis with single cotyledon (MEASC) explant on medium MS+ 6 mg/l BAP+ 0.5 mg/l NAA responded well for shoot induction frequency, multiple shoot induction in Dolichos bean genotype ‘Konkan Bhushan’. The medium MS+ 0.5mg/l BAP+ 0.2 mg/l GA3 was found better for elongation of in vitro regenerated shoots. The genotype ‘Konkan Bhushan’ rooted also earliest in 9.30 days on medium MS+ 1.0 mg/l IBA+ 2% sucrose with highest root induction frequency (79.51%) and maximum roots per shoot (5.04). During hardening, the potting mixture of soil, vermiculite and sand in 1:1:1 showed the highest survival rate.
